Level 2	2.1 Demonstrate use of appropriate sterile technique for handling bacteria. Streak plate. Grow bacteria and transfer bacteria from one plate to another 2.2 Distinguish between bacteria and fungi when they are growing on agar plates and when they are growing on liquid media. 2.3 Examine prepared slides of Mycobacterium tuberculosis the bacterium which causes TB. 2.4 Demonstrate knowledge of the requirements for bacterial growth. (Food, water, space, etc.) and use experimental methods to show how removal of any one of these elements stops bacterial growth. 2.5 Describe the growth requirements for the bacterium which causes tuberculosis. 2.6 Design and conduct an experiment to determine the source of bacteria growing in a flask. Your experimental design must have appropriate controls and a single variable.
Level 3: Complete Level 2 +	3.1 Analyze the results of the experiment done in 2.6 above. State conclusions which can be based on your experimental results. Discuss strengths and weaknesses of your experiment. 3.2 Prepare a bacterial smear; stain and examine with oil immersion lens. Identify bacterial types (cocci,bacilli,etc.) 3.3 Design and conduct an experiment which demonstrates different nutrient requirements of Lac minus and Lac plus strains of E. coli. (Lac minus does not produce the enzyme lactase.)
Level 4: Complete Level 3 +	4.1 Design an experiment to confirm what you learned in 2.6 and 3.1 or to answer a question raised by analysis you did in 3.1. 4.2 Distinguish between encapsulated bacteria and those which are not. Describe the significance of bacterial capsules in disease control, treatment, and prevention.
